Helicoverpa armigera: a global pest

Larvae of the cotton bollworm (Helicoverpa armigera) are dreaded pests all over the world. They have a very wide host range: About 200 different plant species are known as potential food for the voracious insect. The herbivore attacks crops in Africa, South Europe, India, Central Asia, New Zealand, and Australia. Nearly 30% of all globally used insecticides ? Bt toxins as well as pyrethroids ? are applied to protect cotton and other crops against the bollworm.

Resistance to pyrethroids

Pyrethroids are synthetic substances based on compounds of the natural insecticide pyrethrum in Tansy flowers (Tanacetum). They have been successfully applied in fruit, vegetable and crop farming for decades. In cotton bollworms, development of resistance to the particularly effective pyrethroid fenvalerate was observed in Australia since 1983. In 1998, David Heckel, who became director at the Max Planck Institute for Chemical Ecology in 2004, mapped the location of the resistance gene in the genome of the insect for the first time. This gene locus was later found to encode a P450 monooxygenase enzyme. These so-called cytochrome P450 (CYP) enzymes are also known in human medicine, because they can render toxins or pharmaceuticals ineffective by metabolizing the active molecule.

Nicole Jouen, scientist at the institute and expert on P450 enzymes, studied the Helicoverpa armigera strain "TWB" resistant to fenvalerate. She could identify the P450 monooxygenase in this strain, suspected to mediate pyrethroid resistance. After cloning larger genome regions, DNA sequencing, and performing crossing experiments as well as biochemical analyses, she was surprised to find that of the seven P450 enzymes only one, CYP337B3, hydroxylates the fenvalerate molecule to 4'-hydroxyfenvalerate. This chemical reaction increases the resistance to this toxin 42-fold: A LD50 value of 1.9 g of fenvalerate was measured in resistant TWB larvae, whereas half of the non-resistant caterpillars died after intake of only 0.04 g of the toxin.

Unequal Crossing-Over led to the formation of the CYP337B3 Gene

The CYP337B3 gene was formed in a very special way: by a process called "unequal crossing-over" by geneticists. If very similar DNA sequences, for example transposable elements, get in contact with one another during the division of cell nuclei, novel gene combinations occur. As a consequence, some of the genetic information is lost on one DNA strand, and new genetic information is inserted and sometimes even doubled on the other strand. This natural process is important for the evolution of gene families, as could now be observed in the case of the CYP337B3 gene. "For the first time, our results reveal a mutation mediating resistance to an insecticide, which is caused by a crossing-over event," says David Heckel. The scientists studied CYP337B3 and found that the gene consists of parts of two other P450 genes, B1 and B2, encoding enzymes neither of which can detoxify fenvalerate. The unique combination of parts of the B1 and B2 precursor genes in the chimeric B3 gene is responsible for the new capability of the P450 enzyme to bind, hydroxylate, and finally detoxify the insecticide.

Once the CYP337B3 gene was formed, spraying cotton bollworms with pyrethroids caused its frequency in the population to gradually increase. This resistance was successfully delayed in Australia by restricting the use of pyrethroids to once per year, and using other insecticides at other times. This contrasts with the situation in other cotton-growing countries, where pyrethroids were overused, resistance rapidly developed, and then pyrethroids became ineffective against the bollworm. WSJ gets early, slightly uncomfortable look at Google's Project Glass

It's about time someone got the chance to test-drive Google's $1,500 smart-glasses independently, without any rose-colored lenses getting in the way. The Wall Street Journal's Spencer E. Ante just became that lucky person -- he played with prototype specs for 10 minutes and described them as a "wearable smartphone" that weighs just "a few ounces" and is smoothly controlled by voice commands. Saying "OK, Glass" brought up a menu in front of his right eye that made it "easy" to record stills or video, although unfortunately the device wasn't quite ready to show off any phone, messaging or navigation functions.

Overall, Ante acknowledged the "long-term potential" of Project Glass, but in its current form he found the HUD to be "disorienting" and "uncomfortable" -- partly because he instinctively kept closing his left eye to make it all work. Clearly Google still has some work to do if the device is to make a better first impression, but no doubt there's also room for acclimatization on the wearer's part. If the military can get along with this type of eyewear, then hopefully so can everyone else.

Gyms are 'no-go zones' for disabled people, say campaigners

Many local facilities are partially inaccessible and will struggle to cope with increased demand in wake of Paralympics

Many of Britain?s gyms, leisure centres and swimming pools are ?no-go zones? for disabled people and will struggle to cope with an expected surge in interest in fitness activities, following the Paralympic games, campaigners have warned.

A crowdsourced survey of hundreds of gyms across the UK by charity volunteers suggests that many local facilities are partially inaccessible, difficult to navigate and expensive to join. Some did not have specialist gym equipment and nearly half lacked staff trained in disability awareness.

One of the key legacy aims of the Paralympics is to increase participation in sports by Britain?s 11 million disabled people. Just 18% of disabled adults undertake physical activity for more than 30 minutes a week, compared with 38% of non-disabled adults, according to Sport England.

The success of the Games has raised hopes of a surge in interest in disability sports participation, but the survey of gyms and leisure centres, carried out on behalf of the charity Leonard Cheshire Disability, found that many facilities are unprepared to capitalise on the potential demand.

The charity said: ?Gyms and leisure facilities are a crucial ?first point of entry? to sports and physical activity for most adults. Yet for many disabled people they are ?no-go zones?. Inaccessible changing rooms, steps, poor staff training and lack of usable sports equipment are some of the common barriers that can prevent disabled customers from getting involved.?

Guy Parckar, the charity?s head of policy, said that while most gyms were ?broadly accessible? and complied with disability discrimination laws to fit ramps and lifts, many had failed to get right small but crucial details. These could make all the difference when it came to attracting or retaining disabled customers.

Although most gyms provided disabled parking spaces, for example, the survey found that some bays were not wide enough to enable a wheelchair to be unpacked from a car. Parckar said in many cases, improvements such as installing easy-to-read signs or colour contrast markings on steps, would be relatively inexpensive. It was crucial that gyms sought advice from local disabled people on how to improve access, he added.

The survey of 300 UK leisure facilities found that:

? Nearly a third of gyms did not have an automatic door at the entrance to the gym, while some were fitted with heavy internal doors that were difficult to open. One survey reporter said he watched a wheelchair user forced to open such a door by pushing it with his head.

? Although many gyms had lifts, just over one in five were not working properly. A common problem was that lifts were too small for larger wheelchairs and control buttons were set too high. In one case, it was reported that a lift leading to the gym area of a leisure centre did not appear to have been working for more than a year.

? A quarter of swimming pools did not have hoists to support disabled people to get into the water, while 31% of gyms did not have any fitness equipment that was suitable for disabled people.

The survey, carried out as part of Leonard Cheshire Disability?s Exercise Your Right campaign, also highlighted instances where gyms provided excellent services to disabled people. Highly rated gyms were identified as such, not just because they had accessible buildings or equipment but because they practiced good ?customer care and management.? They also employed staff who were trained to be aware of disabled people?s needs.

There is a potentially huge appetite for sport and exercise among disabled people, according to a separate survey of nearly 1,000 disabled people, carried out this week by Leonard Cheshire Disability. It found more than two thirds of respondents said they would like to take part in sport more regularly. Although a quarter said they exercised twice a week or more, a third of those surveyed said they never took part in sport or exercise.

Some 85% of respondents were not members of gyms or leisure centres. Asked why they did not consider joining, more than half said it was too expensive, while 26% said difficulty in accessing facilities put them off

Just under half of respondents said the Paralympics had inspired them to get more involved in sport or exercise, and 26% thought that the Games would encourage more disabled people to take part. But while 89% felt the Games would leave a ?positive legacy? overall, just 12% believed it would lead to improved accessibility.

David Stalker, chief executive of the Health and Fitness Industry Association, which represents more than 3,000 UK gyms and leisure centres, said it was concerned at the findings.

Stalker said the association would be continuing its work with charities to get more disabled people active: ?We will continue to work with our operator members to go above and beyond the minimum requirements to deliver a user friendly environment for all their customers.?

FTC: Anti-bedbug product ads are deceptive

WASHINGTON --?Don't fall for the hype on quick remedies for killing and preventing bed bugs, federal regulators say, as they lodged deceptive advertising charges against two companies marketing anti-bed bug products.

There's no evidence that the ingredients in "Rest Easy" and "Best Yet!," ? including cinnamon, lemongrass and cedar oil ? can eliminate or prevent bed bugs, the Federal Trade Commission said Monday.

"Best Yet!," sold by Cedarcide Industries Inc., also claims to treat head lice. Also deceptive, the FTC says.

The agency sued Cedarcide and RMB Group LLC, marketers of "Rest Easy," in federal court. RMB and its owners agreed in a settlement to make no claims that their product or any pesticide kills or repels bed bugs or creates a barrier against them.

The FTC's case against Cedarcide continues.?

Doctors face manslaughter probe in Morosini death

PESCARA, Italy (AP) -Three doctors are being investigated for alleged manslaughter following the death of Italian footballer Piermario Morosini last season.

Morosini, who was on loan from Serie A side Udinese, collapsed and died during Livorno's Serie B match at Pescara on April 14 after a heart attack. Forensic tests later revealed he suffered from an inherited heart disease.

Both clubs' doctors - Ernesto Sabatini (Pescara) and Manlio Porcellini (Livorno) - are being investigated over their actions, as is Vito Molfese of Pescara's emergency services.

The three doctors were the first to attend to Morosini and have been criticized for not using a defibrillator.

The ambulance was delayed in getting to the player because of a police car blocking its way into the stadium. A window had to be broken so the car could be moved.
